Title: Interakcia perivaskulárneho tukového tkaniva a H2S v mezenterickej artérii v experimente / Interaction of perivascular adipose tissue and H2S in mesenteric artery in experiment

Abstract:

Introduction: Perivascular adipose tissue (PVAT) and hydrogen sulphide (H2S) play important role in modulation of vasoactive responses and may interfere with the etiopathogenesis of essential hypertension. The aim of this study was to evaluate the mutual relationship between PVAT and H2S (endogenously produced and exogenous) in vasoactive responses of isolated mesenteric arteries in adult normotensive (Wistar) and spontaneously hypertensive rats (SHR).
Material and methods: Adult male Wistar and spontaneous hypertensive rats (SHRs) were used. Changes in the isometric tension of MA with or without PVAT were measured after transmural nerve stimulation (TNS), after application of exogenous noradrenaline (NA) and also after exogenously administered H2S. A cystathionine γ-lyase inhibitor, propargylglycine, was used to inhibit endogenous H2S production.
Results: In SHR, hypertension was associated with cardiac hypertrophy and increased arterial contractility, but there were no differences in the amount of retroperitoneal fat between the strains. In SHR, unlike in Wistar rats, PVAT exhibited a pro-contractile effect on contractions mediated by endogenous noradrenaline released from arterial sympathetic nerves. On the other hand, PVAT exhibited the anticontractile effect on exogenous noradrenaline-induced vasoconstriction in both rat strains, but more significantly in SHR. The endogenous H2S produced by both the vascular wall and PVAT demonstrated an anti-contractile effect on the exogenous noradrenaline-induced vasoconstriction in Wistar rats. In SHR, H2S produced by the vascular wall exhibited a pro-contractile effect that was eliminated by the presence of PVAT but not by H2S produced by it. Nevertheless, unlike in Wistar rats, the presence of PVAT potentiated the vasorelaxant effect of exogenously applied H2S in SHR.
Conclusion: Our results confirmed that PVAT of mesenteric artery and endogenously produced H2S exhibit both pro-contractile and anti-contractile properties depending on the type of activated signaling pathway. In SHR, unlike in Wistar rats, the pro-contractile effect of PVAT was associated with perivascular nerve stimulation and, together with the pro-contractile effect of H2S produced by the vascular wall, contributed to pathological changes in SHR. On the other hand, the PVAT of the hypertensive mesenteric artery is endowed with compensatory vasoactive mechanisms, which include both its anticontractile effect and potentiation of the vasorelaxant effect of exogenous H2S. Abstrakt. Úvod: Perivaskulárne tukové tkanivo (PVAT) a sírovodík (H2S) plnia dôležitú úlohu pri modulácii vazoaktívnych odpovedí a môžu byť zapojené do etiopatogenézy esenciálnej hypertenzie. Cieľom tejto štúdie bolo vyhodnotiť vzájomný vzťah medzi PVAT a H2S (endogénne produkovaným a exogénnym) vo vazoaktívnych odpovediach izolovaných mezenterických artérií (MA) u dospelých normotenzných (Wistar) a spontánne hypertenzných potkanov (SHR).
Materiál a metodika: Použili sa dospelé samce Wistar a spontánne hypertenzných potkanov (SHR). Zmeny izometrického napätia MA so zachovaným alebo odstráneným PVAT sa hodnotili po transmurálnej nervovej stimulácii (TNS), po aplikácii exogénneho noradrenalínu (NA) a taktiež po exogénne podanom H2S. Na inhibíciu endogénnej produkcie H2S sa použil inhibítor cystationín y-lyázy, propargylglycín.
Výsledky: U SHR sa hypertenzia spájala so srdcovou hypertrofiou a zvýšenou cievnou kontraktilitou, avšak medzi kmeňmi neboli žiadne rozdiely v množstve retroperitoneálneho tuku. U SHR, na rozdiel od Wistar potkanov, vykazovalo PVAT prokontraktilný účinok na kontrakcie sprostredkované endogénnym noradrenalínom uvoľňovaným z arteriálnych sympatikových nervov. Na druhej strane, PVAT prejavovalo antikontraktilný účinok na vazokonstrikciu vyvolanú exogénnym noradrenalínom u oboch kmeňoch potkanov, ale výraznejšie u SHR. Endogénny H2S produkovaný cievnou stenou aj PVAT preukázal antikontraktilný účinok na vaozokonstrikciu vyvolanú exogénnym noradrenalínom u Wistar potkanov. U SHR vykazoval H2S produkovaný cievnou stenou pro-kontraktilný účinok, ktorý bol síce eliminovaný prítomnosťou PVAT, nie však ním produkovaným H2S. Prítomnosť PVAT zosilňovala vazorelaxačný účinok exogénne aplikovaného H2S u SHR avšak nie u nor- motenzných potkanov.
Záver: Naše výsledky potvrdili, že PVAT mezenterickej artérie a endogénne produkovaný H2S vykazujú prokontraktilné aj antikontraktilné vlastnosti v závislosti od typu aktivovanej signálnej dráhy. U SHR, na rozdiel od potkanov Wistar, je pro-kontraktilný účinok PVAT spojený so stimuláciou perivaskulárnych nervov a spolu s prokontraktilným účinkom H2S produkovaného cievnou stenou prispel k patologickým zmenám SHR. Na druhej strane, PVAT hypertenznej mezenterickej artérie je vybavené kompenzačnými vazoaktívnymi mechanizmami, ktoré zahŕňajú jednak jeho antikontraktilný účinok, ako aj potenciáciu vazorelaxačného účinku exogénneho H2S.
